Losing prime on 03 6.0

I am sure that this topic has been covered but the search feature has left me with no results. My truck known by the ladies in my house as "Leroy" developed a porblem last week in that if it sits more than a day or so it loses its prime. I have checked the ususal suspects, ie the fuel filter housings for a tight fit. Any thoughts out there? I have cranked on it to no avail. though if you hit it with a small shot of ether she perks right up nad runs without any problems. All help and comments appreciated.

here is a bump! my truck will not crank this morning either. it sounds like is is not getting any fuel. it acts like it wants to turn over. strong start but then it just spits. I am in my driveway on a slope with a trailer attached and cannot back out of the driveway into the main road. (would the ether work for my truck?)

I know ether isn't supposed to be used so if you try it don't hose it down or you could blow a head gasket.
