WELCOM TO POWERSTROKE.ORG !!! Pull the door panel on one side or the other, manually lock and unlock the door while watching the rods at the latch assembly and see how they work. The plastic clips that hold the rods in place may be broken or out of adjustment, or you may have the VERY common problem of weak or non-working lock actuators. Happens a lot with those trucks, especially after a couple hundred thousand miles. Lube that latch assembly while you're in there.
