Changing Spartan tunes...
Tomorrow I am planning on reloading my 250 tune and going back through the trans relearn process due to a 2-3 shift problem. This will be the first time I am switching tunes. I need to make sure I do it right because I will be doing it in a remote area (without cell service) in the delta so I don't get hauled off to jail.
 I already have the 250 loaded up. My question is, when I reload the tune do I need to load a different tune first, before reloading the 250 or can I just reload the 250 over itself?

You can just reload the 250 tune.
