Leave the settings alone, that's why you got custom tuning. If you do decide to play with the shift firmness, only increase it by 5% at a times and test drive it for a couple of days before you bump up another 5%.
Or if your not happy how the custom tune runs, call your tune writer up and have them adjust it to your liking.
Also the trans needs time to relearn, usually with in 50 miles.
