Need help Loss of power ???

I am working on this truck. It is not the one in my sig. It is a 2004 6.0L six speed manual with 34,000 miles on it. It drives fine until you shift into overdrive then it develops almost a misfire. It smooths out a little as you get the rpms up above 2400 rpm. It does not seem to do it in the other gears. Any ideas????
